I will try to give an overview.
Dr Behari found that human patients responded better to the nighttime dosing. This was postulated to be because of the circadian rhythm that has us make the most endorphins between 2 and 4 am and that is what we are taking the LDN for -- to make more endorphins. Secondly, when you take the LDN, the LDN blocks the opiod receptors so that other opiods and hormones cannot bind there. Since that means that the body will not feel any endorphins that it has, there is a chance for depression and fatigue during that time. Best to sleep through it rather than having depression during the day. There have been reports of the LDN not working at all with a morning dose.
Having said all of that, there are other reports that morning dosing works just fine. Shrugs. We put the LDN right on our night stand and take it last minute before going to sleep. Most of us with chronic illnesses have terrible sleep patterns already, so for most, taking it at night is the best way. IF after a couple of weeks, there is a problem that needs to be solved, and taking it in the daytime can cure that, then I would give it a try, but not before trying the traditional way that has helped so many and not unless there is a real reason to change the time of the dosing.

Ik neem LDN vlak voor het slapen gaan, dus geen specifieke vaste tijd.
Toen ik met LDN begon had ik wel een vast tijdstip gezien de gewenning aan de ldn en de toenemende klachten.
Wat ik wel merk is dat je niet geestelijk of lichamelijk afhankelijk raakt van dit medicijn ( LDN dus ) en dat heb ik met andere reguliere medicatie wel gehad, dus afhankelijkheid. ( dat valt bij mij ook onder de noemer rotzooi )
en ik kan er alleen maar 1 ding over zeggen, pas ervoor op met wat je slikt met name als je ME hebt. Het komt ( allemaal ) niet ten goede van je klachten.
Nogmaals, dankzij LDN functioneer ik weer en ik kan het echt aanraden.

Simpel uitgelegd:
LDN stimuleert de aanmaak van opiaat receptoren. Deze zorgen dat je meer endorfinen gaat aanmaken.
Oftewel: je gaat alle dagen een beetje sporten. Op den duur gaat je conditie vooruit. (onder normale omstandigheden)
Nog eentje: Je breit alle dagen een paar steken. Na een tijdje heb je een sok. (voor de liefhebbers)
Ik begrijp niet hoe je van extra endorfinen een depressie kan krijgen.
Het enige dat er af en toe gebeurt, is dat het lichaam zich aanpast aan het nieuwe aantal opiaatreceptoren/endorfinen en dat je tijdelijk een euforisch gevoel kan krijgen. Dat zwakt daarna weer af omdat het lichaam zijn normaal evenwicht weer heeft gevonden. Dat is geen depressie waar je dan insukkelt, verre van.
Wat je wel moet weten, is dat het geen kattepis is. LDN helpt je immuunsysteem weer zijn taken op te nemen. Als je belast bent met allerlei kwalen, beestjes, ... dan kan je het gevoel hebben achteruit te gaan ipv vooruit. Echter allerlei klachten verdwijnen tijdens het gebruik wat moed geeft om door te gaan.

Maar ik krijg de indruk (kan het niet helemaal goed verstaan) dat de Meirleir hel alleen over de blokkade heeft?? en niet over het effect dat daarna optreedt?? de aanmaak van extra endorfine??
jammergenoeg eindigt het filmpje te snel..het is me dus niet duidelijk wat hij bedoelt..
Over het algemeen worden mensen juist minder depressief door LDN

TNI Acquires Bihari's Patents; Phase III Trial Planned
Dr. Nicholas Plotnikoff, Chairman of TNI BioTech, recently announced that his company has acquired all of Dr. Bernard Bihari's use-patents for low dose naltrexone. This will enable TNI to proceed with its plans for a phase III clinical trial within the next 12 months, as well as plans to manufacture and sell the drug. Dr. Plotnikoff, quoted on the TNI BioTech website, said:
"I worked with Dr. Bernard Bihari for many years. I believe acquiring all of the technology of Dr. Bihari will allow the company, we hope, to move forward to Phase III clinical trials in both the United States and Africa within the next 12 months. In our opinion, LDN holds great promise for the millions of people worldwide with autoimmune diseases or central nervous system disorders or who face a deadly cancer. It could prove to be the first low-cost, easy to administer, and side-effect-free therapy for cancer, HIV/AIDS, MS and Crohn's disease."
TNI BioTech has begun construction of a large pharmaceutical plant in Managua, Nicaragua for the production of LDN, under the trade name of IRT-103. It expects to be in operation by the end of this month and it is said to be capable of manufacturing well over one billion of the capsules annually. According to their website, the goal of TNI BioTech is "to enable mankind...to combat fatal disease by...mobilizing the body's own immune system using TNI's patented compound(s)".
Dr. Plotnikoff, Emeritus Professor of Pharmacology and Psychoneuroimmunology, College of Pharmacy and College of Medicine, at the University of Illinois in Chicago, holds a PhD in Pharmacology. Prior to TNI, Dr. Plotnikoff helped direct research for Abbott Labs.
[Editor's Note: Some readers have expressed concern that the patenting and manufacture of LDN by a pharmaceutical company would increase the price of LDN for all users. Although a manufactured brand would initially have an increased price, it is probable that the price of LDN as an off-label, compounded drug should not be substantially affected.
